IMMUNOHISTOCHEMICAL STUDY OF PHENOTYPES OF DENDRITIC CELLS IN DENTAL PULPS FROM NON-CARIOUS AND CARIOUS TEETH By Melissa A. Harmon, DDS A thesis submitted in partial fulfillment of the requirements for the degree of Master of Science in Dentistry at Virginia Commonwealth University. Virginia Commonwealth University, 2007 Major Director: Karan J. Replogle, DDS, MS Chair, Department of Endodontics Mature dendritic cells (DCs) in inflamed tissues may promote inflammation but the status of DCs in pulpitis is not known. We hypothesized that DC maturation would correlate with carious lesion depth and that CD4+ cells would be found in association with mature DCs. Pulps were collected from teeth exhibiting: (I) no caries (n=9), (II) shallow dentinal caries (n=5), and (III) deep caries (n=9). Pulpal tissues were cryo-sectioned and positive cells were examined with immunohistochemistry, Mature DCs (CD83+) were almost exclusively restricted to pulps from deep caries. Furthermore, CD209+ DCs in deep

Pulp and plaque microbiotas of children with severe early childhood caries

BACKGROUND AND OBJECTIVE Bacterial invasion into pulps of primary teeth can lead to infection and premature tooth loss in children. This pilot study aimed to explore whether the microbiota of carious exposures of dental pulps resembles that of carious dentin or that of infected root canals. DESIGN Children with severe early childhood caries were studied. Children were consented and extent of ...

Analysis of Trace Elements in Teeth by ICP-MS: Implications for Caries

Teeth are good indicators of environmental exposure to heavy metals and of nutritional status. Inductively coupled plasma-mass spectrometry (ICP-MS) was used to compare the content of trace elements in primary and permanent teeth. For this purpose, primary teeth were collected from 64 children and 112 permanent teeth were collected from 40to 60-year-old adults. The data were assessed statistica...

Detection and Characterization of Human Teeth Caries Using 2D Correlation Raman Spectroscopy

Background: Carious lesions are formed by a complex process of chemical interaction between dental enamel and its environment. They can cause cavities and pain, and are expensive to fix. It is hard to characterize in vivo as a result of environment factors and remineralization by ions in the oral cavity. Objectives: The development of a technique that gives early diagnosis which is non-invasi...

Dental Pulp Cells Isolated from Teeth with Superficial Caries Retain an Inflammatory Phenotype and Display an Enhanced Matrix Mineralization Potential

We have isolated dental pulp cells (DPCs) from three healthy (hDPCs) and three carious (cDPCs) donors and shown that compared to hDPCs cells isolated from superficial carious lesions show higher clonogenic potential; show an equivalent proportion of cells with putative stem cell surface markers; show enhanced matrix mineralization capability; have enhanced angiogenic marker expression and retai...

Relation between pulpal neuropeptides and dental caries

INTRODUCTION Dental pulp has neural fibers that produce neuropeptides like Substance P (SP) and calcitonin gene-related peptide (CGRP). The inflammation of dental pulp can lead to an increase amount of SP and CGRP release, especially in symptomatic irreversible pulpitis. Therefore, it can be assumed that neuropeptides have some role in the progression of inflammation of the dental pulp. The aim...
